Her original first name was spelled "Lucy," but she changed the spelling during her teenage years as a form of personal rebellion. Her parents named their daughters with initials matching their own—Lyndon Baines Johnson and Lady Bird Johnson—resulting in the initials LBJ for both daughters.

Johnson was raised Episcopalian. Her father was a member of the Christian Church (Disciples of Christ), while her mother converted to the Episcopal Church. At age 18, Johnson converted to Roman Catholicism through a conditional baptism; she had been baptized as an infant with water and in the name of the Trinity by an Episcopal priest in Austin, Texas. This rebaptism at age 18 attracted media attention due to differences in doctrinal requirements between denominations.

At age 16, Johnson was present in Washington, D.C., during the assassination of President John F. Kennedy on November 22, 1963. She was attending a Spanish class at the National Cathedral School at the time. She did not initially know if her father had been injured but noticed the presence of Secret Service agents on her school campus shortly afterward, indicating he had been sworn in as President.

Johnson briefly attended Georgetown University School of Nursing and Health Studies but left in 1966 due to the school's policy against married students, as she married her first husband that same year. In August 1966, she learned to cook from Zephyr Wright, who served as the White House chef during her family's administration.

In her professional career, Johnson has held several leadership roles. Since 1993, she has been the chairman of the board and manager of LBJ Asset Management Partners, a family office. She also co-founded and served as chairman of BusinesSuites, a company overseeing executive office suites, with her second husband in 1989. She earned a Bachelor of Liberal Studies in Communication from St. Edward's University in 1997.

Her involvement in civic and philanthropic activities includes serving on the board of the LBJ Foundation, fundraising for the Lady Bird Johnson Wildflower Center and the American Heart Association, acting as a trustee of Boston University, and serving on the advisory board of the Center for Battered Women.

Johnson married Patrick John Nugent on August 6, 1966. The marriage was televised and appeared on the cover of Life magazine. The couple divorced, and the marriage was annulled by the Catholic Church in August 1979. She married Ian J. Turpin, a Scottish-born Canadian financier and president of LBJ Asset Management Partners, on March 3, 1984. Through this marriage, she has a stepson.

In April 2010, Johnson was diagnosed with Guillain–Barré syndrome, an autoimmune disorder affecting the peripheral nervous system. She was treated at the Mayo Clinic in Rochester, Minnesota, and subsequently returned to Austin in May 2010. Her healthcare providers described her case as less severe than typical, and she experienced a full recovery.
