Hancock Lee was born in 1653, the son of Richard Lee I, Esq. His date of death is recorded as May 25, 1709. Lee was involved in colonial governance and judiciary roles within Virginia.

In 1677, Hancock Lee served as a justice in Northampton County. He later relocated to Northumberland County, where he served as a justice in 1687. His political career included membership in the Virginia House of Burgesses during the years 1688 and 1689.

Lee was married twice. His first marriage was to Mary Kendall, the daughter of William Kendall. His second marriage was to Sarah Allerton, the daughter of Isaac Allerton Jr., Esq. Through his marriage to Sarah Allerton, Lee was the father of Elizabeth Lee. Elizabeth Lee was the paternal grandmother of the 12th President of the United States, Zachary Taylor.

He owned multiple estates, including one named Ditchley, located in Northumberland County, Virginia.
