John Holles is the son of John Holles, 2nd Earl of Clare, an English politician and peer. He is also the grandson of Gilbert Holles, 3rd Earl of Clare, an English politician. His mother is Lady Grace Holles, who was the daughter of Gilbert Holles. Lady Grace Holles died in 1700. Thomas Pelham-Holles, the 1st Duke of Newcastle-upon-Tyne, is his maternal great-grandson and served as Prime Minister of Great Britain between 1693 and 1768.
